粒線體動態平衡在粒線體的功能和細胞遷移中扮演重要角色。先前的研究指出瘦長型的粒線體會有較高的粒線體膜電位和較好的粒線體呼吸作用。另外,瘦長型的粒線體對於細胞遷移具有抑制作用。相反的,碎片型的粒線體則具有較低的粒線體膜電位,並且有較差的粒線體呼吸作用。另外,碎片型的粒線體也被發現對細胞遷移有促進作用。粒線體動態平衡和細胞遷移都會受到肌動蛋白的動態平衡影響。人類線苷酸環化酶關聯蛋白質二號會聚集球狀肌動蛋白,並且破壞肌動蛋白絲的結構。然而,目前為止尚未有研究指出人類線苷酸環化酶關聯蛋白質二號在癌細胞中和粒線體動態平衡及細胞遷移之間有關連。在我的研究當中發現大量表現人類線苷酸環化酶關聯蛋白質二號在實驗用增殖表皮子宮頸癌細胞 (HeLa) 和胃癌細胞 (SC-M1) 中時,會影響粒線體動態平衡而導致粒線體的型態和功能改變。另一方面,人類線苷酸環化酶關聯蛋白質二號的大量表現會擾亂肌動蛋白絲的型態並且降低細胞遷移的能力。
Mitochondrial dynamics is important for mitochondrial functions and cell migration in cancer cells. Previous studies showed that elongated mitochondria elevate mitochondrial membrane potential, enhance mitochondrial respiration, and decrease cell migration; fragmented mitochondria decrease mitochondrial membrane potential, reduce mitochondrial respiration, and promote cell migration. CAP2 sequesters G-actin and disrupts actin filaments. Moreover, CAP2 is up-regulated in hepatocellular carcinoma, and the knockdown of CAP2 suppresses the cell migration. However, there is no study discussing the correlation of CAP2 with both mitochondrial dynamics and cell migration in cancer cells. In this thesis, I found that CAP2 overexpression affected mitochondrial dynamics, and caused mitochondrial morphology and functions were changes in both HeLa and SC-M1 cancer cells. In addition, CAP2 overexpression disturbed actin filaments and decreased the activity of cell migration.
