蛋白質水解酵素為所有酵素中使用最廣的一種,可使用於清潔劑產業、啤酒釀造、烘培工業及乳品加工等產業,而利用來自天然生物體的幾丁聚醣固定蛋白水解酵素,可節省生產成本。本研究利用不同乙醯化程度之幾丁聚醣粉末依不同方法製備幾丁聚醣顆粒,進行蛋白水解酵素Protease A之固定,探討製作顆粒的方法(烘乾、濕潤及凍乾)、幾丁聚醣擔體之乙醯化程度(乙醯化程度5%和10%,在本論文裡以DA 5&DA 10表示) 及固定化方法對固定化之Protease A活性的影響。
首先,實驗結果發現凍乾的顆粒大小和濕潤的差不多,不過由於凍乾顆粒的表面會形成多孔洞,所以固定的酵素活性最高,其次是濕潤,最差的是烘乾的顆粒。至於在不同乙醯化程度的幾丁聚醣顆粒方面,低的乙醯化程度固定的酵素活性較高。
再者,以新的交聯吸附固定法來固定酵素,可得到比共價固定法更高的固定量、活性表現和比活性,不過在重複使用性方面則是劣於共價固定法,但若以經濟層面進行考量,交聯吸附固定法應用於酵素固定化具有很大的優勢。
最後,酵素因為有擔體的保護,所以對於溫度和pH值的變化其穩定度明顯提高,較適反應溫度和pH也變寬。在重複使用性方面,以DA 5的濕潤擔體而言,經共價法固定的酵素可重複使用20次而仍保有70%以上的相對活性。經由計算後得到的酵素動力學參數Km和Vmax,固定化酵素的Km和游離酵素差不多,但是其Vmax則是小於游離酵素。
