多重影像系統之應用結合了原本屬於各種影像系統之特色及優點,大幅改善影像結果的解析度及功能性,增加臨床上之應用價值。具有多重影像報導基因之動物模型,更能同時運用多種成像系統進行觀測並互相驗證。在本篇論文裡,我們成功結合了三種不同影像報導基因,包括利用螢火蟲冷光酵素、綠螢光蛋白、以及第一型單純疱疹病毒胸腺嘧啶激酶,並且利用專一於腦部表達之纖維母細胞生長因子-1.B 啟動子來驅動轉殖基因。利用各成像系統相對應之探針,可以檢視所生產出基因轉殖動物之基因表達模式。我們建立出帶有此FGF-1.B-TMIR 載體之基因轉殖小鼠模型後,希望可以利用其影像上之優勢,應用於追蹤腦部細胞之增生、分化、遷移、以及發展之機制及過程。
我們檢視了由540 鹼基對長度之纖維母細胞生長因子-1.B 啟動子驅動之基因轉殖鼠,可以成功地表達後續之三種影像報導基因。也於體外細胞實驗中,利用暫態轉染TMIR 載體之方法來確認各報導基因之功能無誤。目前成功保持六個獨立F1B-TMIR 之基因轉殖鼠品系,各攜帶不同套數之轉殖基因。由成鼠活體冷光影像實驗之結果說明,此轉殖基因如預期地集中於腦部。但我們也發現在不同品系之間其基因表現模式稍有差異。造成差異之原因可能來自於不同品系之間所攜帶不同之轉殖基因套數,以及轉殖基因嵌插入染色體位置不同所致。在某些品系中,轉殖基因的表現主要在前腦腹側以及嗅覺系統,而其餘之品系主要分佈於腦幹以及頭骨軟骨組織。而實驗結果顯示,轉殖基因的表現在胚胎發育過程較為廣泛,如胚胎及幼鼠之軟骨系統,包括耳廓及耳道等處、軟骨膜、黏膜組織以及嗅覺系統等,而在成鼠之表現分布則較為集中。我們需要對胚胎及成鼠組織以組織免疫染色進行觀察及比較,才能確認幼鼠與成鼠表現模式之差異。具有多項成像系統報導基因之小鼠將有助於對纖維母細胞生長因子-1 在胚胎發育及各種疾病過程中扮演角色之探討。
