  • 類胡蘿蔔是黃色菊花的主要色素,然而目前菊花標本的保色方法,主要以酸性水溶液浸泡菊花標本,長久浸泡後有花色變淡、色素滲出、花瓣癱軟等缺點。本次實驗以黃秀鳳菊花(Chrysanthemum sp.)做為研究材料,使用含不同有機酸之非極性溶液浸泡菊花標本。結果顯示,菊花浸泡於分別含4種不同有機酸之保存液(0.25% citric acid 、0.25% boric acid、0.25% propanoic acid 和
    0.25% acetic acid )後,外觀形態會稍微萎縮,其中以0.25% citric acid和0.25% boric acid 保存液對菊花的黃色保存效果好,色素滲出不明顯。
    吸光值測定結果顯示,菊花浸泡於保存液30天後,其類胡蘿蔔素含量確實會隨著浸泡時間越長而下降;浸泡於保存液0.25% citric acid 、0.25% boric acid、0.25% propanoic acid 和0.25%
    acetic acid後,類胡蘿蔔素濃度含量下降率分別為:26.75% 、17.63%、62.02%、65.03%。若將保存液處理過的菊花包埋於環氧樹脂則可延長其顏色的保存。HPLC分析結果顯示,菊花浸泡保存液後,會造成類胡蘿蔔素訊號波峰位移、消失或增加,表示保存液可能會破壞原本菊花色素種類。而徒手切片觀察新鮮與浸泡保存液後的菊花花瓣,發現原本在新鮮花瓣中,類胡蘿蔔素被有色體包覆呈現小顆粒狀平均分布於細胞內;以保存液浸泡過的花瓣,有色體會被破壞導致類胡蘿蔔素滲出。
    本次實驗所用的保存配方,雖然會造成類胡蘿素含量下降,但與現在普遍使用的水溶液配方相較,能較有效保存顏色而形態雖稍有萎縮,但不致癱軟。


    Carotenoids are the main pigments of yellow chrysanthemums. In the past people used acid solution for yellow color preservation but with time went by the treated
    chrysanthemums lost their color and became shrank obveriously. For better preservation, we use 4 different mixture of organic solution and test their ability to
    preserve yellow color of Chrysanthemum sp. The 4 organic solution only different in their weak acid compound, 0.25 % citric acid, 0.25 % boric acid, 0.25 % propanoicacid, 0.25 % boric acid respectively. Organic mixtures containing 0.25 % citric acid or 0.25% boric acid have better preservation on color of chrysanthemum. compared to those immerse in acid solution with little carotenoids leakage, and petals shrank slightly. However, light wavelength absorption test and HPLC indicate that the four organic mixtures did decrease content of carotenoids and isomerize its structure. Hand-cutting section reveals that chromoplasts which originally distribute in cells break out so that carotenoids spreads around in cytosol when chrysanthemums are immersed in the four organic mixtures. If chrysanthemums which are immersed in organic mixtures previously are embedded in Epoxy, the yellow color can be
    preserved for a longer time. The prescription of our conservation method although caused a decreasing in caroteniods, but to compare those common aqueous solution, it showed a more effective color conservation and morphology that can highly rescue from wither and limped phenomena.
