  • 蘇力桿菌是一種能形成孢子的革蘭氏陽性(Gram positive)好氣性桿菌,菌體兩端鈍圓,有些種類菌體周生鞭毛,具有運動能力,然而有些菌種不生鞭毛,因此不具運動能力。蘇力桿菌通常2~8個呈鏈狀,廣泛地存在於土壤之中[Yu, 1990]。到目前為止,至少已知有38種此類菌株可產生出殺蟲性毒素。我們的實驗目的就是在探討運動性在蘇力菌生理上扮演的角色,在實驗過程中,我們定義出一個物理量〝平均運動速率〞(average motile rate),來表示菌體在生長過程中的運動情形。
    首先,我們藉由觀察平均運動速率與菌體量、蘇力菌素產量隨環境因子的變化趨勢,來探討平均運動速率與蘇力菌生理之間的關連性。根據實驗結果,可得到以下結論:

    (1) 作為觀察菌體生長的新資訊

    (2) 預測蘇力菌素的產量

    其次我們想要進一步瞭解,不同的平均運動速率時期,對菌體而言是否代表了不同的生理時期。因此我們選擇了運動速率的高峰期、遞減期與平穩期,將通氣量、溫度與酸鹼值做一為時二十分鐘的脈衝變化,根據實驗結果可得到結論如下:

    (1) 對蘇力菌而言,當批次系統在醱酵前期受到一定程度的干擾時,對於產物的生成都是不利的

    (2) 當平均運動速率愈高時,環境的改變對菌體的生理影響愈大

    (3) 傳統的指數生長期可藉由菌體平均運動速率,再區分成不同生理活性時期


    Bacillus thuringiensis is an aerobic, Gram positive bacterium which produce spores when the environment goes against its growth. Some sorts of Bacillus thuringiensis have surrounding flagella while the others not. Those who possess flagella have the ability to swim. Bacillus thuringiensis cells usually gather in the form of chains while each chain is composed of 2-8 cells; they are frequently found in the soil. At least 38 strains are known to produce insecticides. This work is to study the role of the motility which plays in the physiology of Bacillus thuringiensis. We define a term "Average Motile Rate" (AMR) to describe the motivation of cells during the cultivation.
    The changing tendencies of AMR, biomass and yield of thuringiensin which respect to some environmental factors are observed to investigate the relationship between the AMR and the physiology of Bacillus thuringiensis. The results showed that:

    (1) The AMR gave new information of cell growth

    (2) The AMR could aid prediction of the yield of thuringiensin

    Also, it is desired to know whether each AMR period indicates a

    corresponding physiological period of Bacillus thuringiensis or not. Pulses of aeration rate, temperature and pH value which last 20 minutes were carried out during the peak period, the decay period and the stable period of the AMR. We then draw following conclusions:

    (1) It was harmful to the production of thuringiensin when the cultivation was disturbed during its early fermentation stage

    (2) The influence upon the cell physiology increased with higher values of AMR

    (3) The conventional exponential phase could be further split into two physiological active stages according to the AMR
