系統生物學為現今學術重點方向之ㄧ,把生物系統化去深度了解其內部組成結構,無論對發炎反應、傳染病、甚至是癌症的醫療都有很大的突破。而核心網路(Core-Network)為系統生物學研究重點之一,分辨核心網路不僅可以簡化複雜反應的網路結構且不失其網路結構重要性,更可以在分析核心網路過程中發現一些重要的生物資訊等。近幾年來,許多文獻中也提及核心網路的重要性與其辨識方法,而本論文是利用參數敏感度的觀念,利用統計方法MPCA(Multiway Principal Component Analysis)一層一層去分辨核心網路,此方法能將所有蛋白質、時間點、反應式全部一次分析處理,改善了以往分析核心網路耗時的缺點,且分析結果準確性也很高。
而本論文研究重點為以發炎反應(1) 細胞質IKK刺激至細胞核轉錄因子NFkB;(2) cytokine TNFα刺激至細胞核內轉錄因子NFkBn,兩個完整網路結構為例子,試圖利用MPCA(Multiway Principal Component Analysis)去分辨其核心網路。而論文內容大綱為敘述MPCA分辨核心網路處理方法與處理過程中得到一些生物資訊之相關討論。
