  • 雙酚A (bisphenol A, BPA)為塑膠製品中常用的塑化劑,能與雌激素受體鍵結,並為一內分泌干擾物質。BPA為脂溶性,具生物累積性,會囤積在脂肪細胞。脂肪細胞(adipose cells)會分泌脂肪激素(adipokines),如脂瘦素(leptin)和脂聯素(adiponectin),以來維持生物體內能量代謝的恆定性。若BPA累積在脂肪細胞時,可能會影響脂肪激素之調控而誘發代謝性疾病。因此,本研究目的為調查妊娠婦女、不孕症婦女和臍帶血中BPA濃度,以及探討BPA對脂肪激素和雌激素受體基因表現之影響。
    由高效能液相層析儀(high-performance liquid chromatography)結合紫外光偵測儀(ultraviolet detector)分析結果發現,妊娠婦女血漿中BPA濃度略高於不孕症婦女,分別為5.44 □ 6.04 ng/ml和5.20 □ 5.30 ng/ml (平均值□標準差; p = 0.784)。除此之外,臍帶血中能偵測到BPA (1.06 □ 2.39 ng/ml),表示胎兒會經由胎盤暴露到BPA,並且妊娠婦女血漿中BPA濃度與胎兒體重呈顯著負相關(r = -0.304, p = 0.010)。另外,分別以定量聚合酶鏈鎖反應(real-time polymerase chain reaction)分析組織中脂肪激素和雌激素受體基因表現,及酵素連結免疫法(enzyme-linked immunosorbent assay, ELISA)分析血漿中脂肪激素蛋白質濃度。結果顯示,妊娠婦女血漿中BPA濃度,與胎盤組織之雌激素受體□ (r = 0.230, p = 0.048)及雌激素受體□ (r = 0.168, p = 0.150)基因表現量呈正相關。此外,妊娠婦女血漿中BPA濃度與血漿中脂瘦素(r = 0.068, p = 0.563)和脂聯素(r = 0.015, p = 0.899)濃度呈正相關,但與胎盤組織之脂瘦素(r = -0.060, p = 0.612)和脂聯素(r = -0.027, p = 0.824)基因表現量呈負相關。
    由上述結果可知,胎兒在母親懷孕期間會經由胎盤暴露到BPA,且BPA暴露會影響胎兒之出生體重。BPA暴露會影響雌激素受體基因表現量,因此BPA對人體之不良健康效應受關注。
