正常細胞轉變成癌細胞主要和基因的調控和蛋白質的表現產生改變有關。本實驗的研究目的是比較正常乳腺細胞(MCF-10A)、非侵略性乳癌細胞(MCF-7)和侵略性乳癌細胞(MDA-MB-231)之間表現蛋白質的差異,試圖找出和腫瘤發生及轉移有關的蛋白質。利用二維差異電泳以及MALDI-TOF MS分析共鑑定出112個蛋白質在正常乳腺細胞(MCF-10A)、非侵略性乳癌細胞(MCF-7)和侵略性乳癌細胞(MDA-MB-231)這三株細胞中具有差異表現,其中有38個蛋白質(如:GDI 2)未發現和乳癌有相關。在所有鑑定到的蛋白質當中,有27個蛋白質推測和腫瘤的發生有關(如:Eukaryotic translation initiation factor 5A-1),21個蛋白質和癌細胞的轉移有關(如:NAD(P)H dehydrogenase [quinone] 1)。另外,使用西方墨點法再次確認部分蛋白質的表現量。總結來說,本實驗鑑定到許多不同階段乳癌相關的蛋白質,這些研究結果可能有助於往後發展為臨床上診斷和治療乳癌的生物標誌分子。
在哺乳動物中,性別的發展是由基因和荷爾蒙所調控,始於受精卵的染色體結構(XY或XX)。接著由Y染色體上的SRY基因的表現而促使性腺的發育,形成男性的睪丸。本實驗利用蛋白質體學為基礎的方法,包括二維差異電泳和MALDI-TOF MS,分析羊水中的男性和女性胎兒細胞之間表現差異的蛋白質。在三次獨立二維差異電泳分析後,共有46個蛋白質點表現量有差異,分別屬30個不同的蛋白質,其中有7個蛋白質(annexin A1, cathepsin D, cytoskeletal 19, protein disulfide-isomerase A3, F-actin-capping protein subunit beta, Rho GDP-dissociation inhibitor 1 and vimentin)在三次獨立實驗的其中兩次實驗當中都顯示為表現差異1.5倍以上,其中一些蛋白質使用西方墨點法做再次的驗證。本實驗所鑑定的差異表現蛋白質可能可以做為男女性鑑別的生物標誌分子,在懷孕早期從母親陰道分泌物中偵測。此外,本研究方法所建立的實驗平台可以提供往後尋找與染色體異常的胎兒相關的生物標誌分子。
