心肌梗塞為冠狀動脈硬化,導致心肌組織產生缺氧壞死。臨床上可以利用人工補綴片來修補壞損的心肌組織 ,以恢復心臟的部分功能。本篇研究利用甲基纖維素培養系統,製作出心肌細胞片並嵌入多孔性細胞支架中,體外培養成新穎性組織工程細胞支架。在實驗的一開始,利用甲基纖維素培養系統,成功製作出不貼附於培養皿的胚胎體。並加入適量濃度的維他命C (10-2mol/l) ,並以流式細胞儀分析,在加入維他命C誘導的條件下,分化第8 天後大約有15 % 左右的細胞帶有著心肌細胞的特有蛋白 myosin heavy chain (MHC)。在成功分化出心肌細胞後,利用甲基纖維素溫度敏感性的特性得到具有跳動性質的細胞片,並將細胞片置入製備好的多孔性牛心包膜細胞支架的切口中,進行體外培養,探討細胞在其內增生、分化的情形。製作出三明治細胞支架後,為了驗證可行性,藉由切片染色H&E 觀察細胞是否成功坐落於心包膜切口中。接著更近一步運用免疫螢光染色,對坐落於心包膜中的細胞和細胞片進行心肌細胞特有蛋白( myosin heavy chain,connexin 43,α-actinin) 辨認,証實置入進心包膜的細胞確實包含著心肌細胞。接著,藉由免疫螢光染色觀察,在細胞片中細胞支架孔洞裡有新生的細胞外間質蛋白(fibronectin,collagen I,collagen III),證實了直接植入細胞片的方法有利於細胞在牛心包膜中環境生長。然而,在實驗的進行中。藉由免疫染色觀察到細胞支架內有著血管組織的產生,顯示著胚胎幹細胞在分化時,不只是分化成心肌細胞於細胞支架內,同時也分化出提供心肌細胞養分的血管產生。以上結果顯示以胚胎幹細胞製作組織工程細胞支架是具有可行性的。
