秋水仙胺,為一種細胞分裂的抑制劑,它會加成受紫外線照射的中國倉鼠卵巢細胞死亡。在本研究中,秋水仙胺被發現具有抑制受紫外線處理的細胞DNA修復能力。這個秋水仙胺所造成的現象,可以被三種不同的過程所觀察到。例如:細胞核大小分析(Nucleoid analysis),慧星檢測法(Comet assay),PCR定量法(QPCR)。然而,本研究中又觀察到秋水仙胺同樣可以使經過氨化鉑處理的細胞加成性死亡,但是這種情況並不發生在X-ray 所照射細胞上。秋水仙胺可能專一的抑制核甘酸切除修復(Nucleotide excision repair)。本篇研究推測應用不同的作用機制來共同處理藥物,可增加癌症化學治療的效果。
Colcemid, a mitotic inhibitor, promotes UV-induced apoptosis in CHO-K1 cells. In this study, colcemid was found to inhibit DNA repair in the UV-irradiated cells. This effect of colcemid was observed in three different procedures, i.e. The nucleoid size analysis, the comet assay and quantitative polymerase chain reaction. Since colcemid promoted apoptosis in UV-irradiated or cisplatin but not X-ray irradiated treated cells, this agent might specifically inhibit the nucleotide excision repair. This study suggests that effective cancer chemotherapy can be achieved by the combination of drugs with different action mechanisms.
